Overview

RetroMAD1 is a 41.3 kDa chimeric recombinant fusion protein developed by Biovalence Technologies as a broad-spectrum antiviral agent. The molecule is composed of three distinct antimicrobial proteins: Retrocyclin-101 (a synthetic theta-defensin), MAP-30 (Momordica anti-HIV protein), and Dermaseptin-S1 (an antimicrobial peptide), which are linked together via peptide linkers. Originally developed for veterinary use against Feline Leukemia Virus (FeLV), RetroMAD1 has been repurposed for the treatment of COVID-19. Its primary mechanism of action involves the inhibition of the SARS-CoV-2 3CL protease (3CLpro), an essential enzyme for viral polyprotein processing and replication. In vitro studies have demonstrated that RetroMAD1 effectively prevents SARS-CoV-2 infection in human bronchial cells with potency comparable to remdesivir and without significant cytotoxicity. It is designed for oral administration.
