RetroVirox is developing small molecule inhibitors that target the HIV Nef protein to prevent the downmodulation of MHC class I (MHC-I) molecules on the surface of infected T cells. By blocking this Nef-mediated immune evasion mechanism, the inhibitors restore the presentation of viral antigens, allowing cytotoxic T lymphocytes (CTLs) to recognize and kill HIV-infected cells. This therapeutic approach is designed to be a critical 'kill' component of 'kick and kill' HIV eradication strategies, aimed at eliminating the latent viral reservoir. The program is currently in the preclinical stage, with lead compounds demonstrating high potency and selectivity.
