Retrovirus vector LN is a specific Moloney murine leukemia virus (MoMLV)-based retroviral vector used primarily for gene marking in clinical research settings, such as those conducted at the Masonic Cancer Center, University of Minnesota. The vector typically carries the neomycin phosphotransferase (neo) gene, which serves as a selectable marker. In the context of autologous peripheral blood stem cell (PBSC) transplantation for non-Hodgkin lymphoma and Hodgkin lymphoma, a portion of the patient's harvested progenitor cells is transduced with the LN vector ex vivo. These marked cells are then re-infused following high-dose chemotherapy and/or radiation. This technique enables investigators to monitor the long-term engraftment and stability of the transplanted cells and helps determine whether disease recurrence arises from contaminating tumor cells within the graft or from endogenous cells that survived the conditioning regimen.
