Overview

Rev CD123-5B9 is a bispecific antibody adaptor molecule, specifically a target module (RevTM), developed as part of the **Revocable Chimeric Antigen Receptor (RevCAR)** platform. It is designed to treat **acute myeloid leukemia (AML)** by bridging RevCAR-engineered T-cells and malignant cells expressing **CD123** (interleukin-3 receptor alpha chain). The molecule consists of two single-chain fragment variable (scFv) domains: one specific for the **E5B9 peptide epitope** (derived from the human nuclear protein La/SS-B) expressed on the RevCAR T-cell, and another specific for CD123, which is overexpressed on AML blasts. This modular approach allows for controllable CAR T-cell therapy; the engineered T-cells are only active in the presence of the RevTM adaptor, providing a safety switch to mitigate potential toxicities like cytokine release syndrome by discontinuing the administration of the short-lived adaptor molecule.
