Drug intelligence / Profile preview
REV-ERB modulator is a preclinical-stage small molecule agonist of the REV-ERB nuclear receptors (REV-ERBα and REV-ERBβ) being developed by Pelagos Pharma. Functioning as an exercise mimetic, the compound is designed to activate the same beneficial metabolic, mitochondrial, and transcriptional pathways as aerobic exercise. Pelagos Pharma is developing the therapeutic for the treatment of heart failure, obesity, sarcopenia, and cognitive decline (including neurodegenerative diseases such as frontotemporal dementia and Alzheimer's disease). As of 2026, the program is in the lead optimization phase, with development candidate nomination projected for Q3 2027 and IND-enabling studies projected for 2028.
