Revaprazan + itopride is a fixed-dose oral combination used primarily in the management of functional dyspepsia and potentially other acid-related gastrointestinal disorders. Revaprazan is a selective potassium-competitive acid blocker (P-CAB) that reversibly inhibits the gastric H(+)/K(+)-ATPase enzyme (proton pump), resulting in reduced gastric acid secretion and rapid increase in intragastric pH. Itopride is a prokinetic agent acting as both a dopamine D2 receptor antagonist and an acetylcholinesterase inhibitor, thereby increasing acetylcholine concentrations at enteric nerve endings, which in turn enhances gastric motility, increases lower esophageal sphincter pressure, accelerates gastric emptying, and improves gastro-duodenal coordination. The combination is designed to provide symptomatic relief from functional dyspepsia by targeting both impaired motility and excessive acid secretion. In clinical studies, the combination has demonstrated bioequivalence and a favorable tolerability profile compared to individual monotherapies, with no significant pharmacokinetic drug-drug interaction[1][3][5][7].
