Overview

RevCAR-E7B6 T-cells are an engineered cell therapy utilizing the **Reverse Chimeric Antigen Receptor (RevCAR)** platform, designed to enhance the safety and controllability of CAR-T cell treatments. Unlike conventional CAR-T cells that express an antibody-derived binding domain (scFv), RevCAR T-cells express a small, non-immunogenic peptide epitope called **E7B6**, which is derived from the human nuclear protein La/SS-B. These T-cells are therapeutically inactive on their own; they require the administration of a bispecific adapter molecule known as a **RevTM (Target Module)**. The RevTM bridges the RevCAR T-cell (by binding the E7B6 epitope) to a specific tumor-associated antigen (such as CEA or GD2), thereby triggering T-cell activation and tumor lysis. This binary system allows for "on/off" switchability and dose-titration of T-cell activity by controlling the administration of the RevTM, potentially reducing the risk of severe adverse effects like cytokine release syndrome.
