RevM10 is a dominant-negative mutant of the HIV-1 Rev protein, designed as a gene therapy approach to inhibit HIV replication. The Rev protein is essential for the nuclear export of unspliced and singly spliced viral mRNAs in HIV-1, which are required for producing structural proteins and enzymes necessary for viral replication. RevM10 contains mutations that disrupt its nuclear export signal (NES), allowing it to bind the Rev response element (RRE) on viral RNA but preventing proper interaction with cellular export machinery. This competitive inhibition blocks wild-type Rev function, thereby inhibiting production of infectious virus particles. RevM10 has been studied primarily as a gene transfer therapy by introducing its gene into hematopoietic stem/progenitor cells or T cells to confer resistance against HIV infection in their progeny. Clinical trials have explored its use in patients with HIV-related non-Hodgkin lymphoma and other settings[1][2][4][5][6].
