Overview

RevTM CD123-7B6 is a bispecific antibody adaptor molecule developed as part of the RevCAR (Reversible Chimeric Antigen Receptor) platform, primarily for the treatment of acute myeloid leukemia (AML). It consists of two single-chain fragment variable (scFv) domains: one that binds to the E7B6 peptide epitope expressed on RevCAR-modified T-cells, and another that targets the CD123 antigen (Interleukin-3 receptor alpha) on AML blast cells. As a RevCAR Target Module (RevTM), it acts as a molecular bridge, redirecting T-cell cytotoxicity specifically toward CD123-positive malignant cells. This modular design allows for "ON/OFF" control of CAR-T activity by regulating the administration of the RevTM, potentially reducing the risk of cytokine release syndrome and off-tumor toxicity compared to conventional CAR-T therapies. The molecule has shown high potency in preclinical models, effectively lysing AML cells at picomolar concentrations.
