Overview

RevTM CD33-5B9 is a bispecific antibody adaptor molecule, referred to as a RevCAR Target Module (RevTM), developed for use within the modular RevCAR (Reversible CAR) T-cell platform. It is engineered with two single-chain fragment variable (scFv) domains: one specific for the E5B9 peptide epitope, which is part of the extracellular domain of RevCAR-engineered T-cells, and another targeting the CD33 antigen, which is highly expressed on acute myeloid leukemia (AML) blasts. By acting as a molecular bridge, RevTM CD33-5B9 redirects the cytotoxic activity of RevCAR T-cells toward CD33-positive malignant cells. This system provides a safety mechanism where CAR T-cell activity is dependent on the presence and concentration of the RevTM, allowing for reversible ON/OFF control of the therapy. It is currently being investigated as a research compound for the treatment of AML.
