Overview

RevTM CD33-7B6 is a bispecific antibody adaptor molecule, specifically a RevCAR Target Module, designed for the treatment of acute myeloid leukemia (AML). It is a key component of the Reverse Chimeric Antigen Receptor (RevCAR) platform, which aims to enhance the safety and control of CAR-T cell therapies. The molecule consists of two single-chain fragment variables (scFvs): one targeting the CD33 antigen commonly expressed on AML blasts, and the other targeting the E7B6 peptide epitope expressed on RevCAR-modified T-cells. By acting as a bridge, RevTM CD33-7B6 redirects the cytotoxic activity of RevCAR T-cells toward CD33-positive malignant cells. This modular system allows for the reversible regulation of T-cell activity by controlling the administration of the adaptor molecule, potentially reducing off-target toxicities and cytokine release syndrome. Preclinical studies have demonstrated potent, dose-dependent cytotoxic activity against AML cells in the picomolar range.
