Drug intelligence / Profile preview
Revuforj + venetoclax is an investigational combination regimen consisting of the oral menin-KMT2A inhibitor revumenib (Revuforj) and the oral BCL-2 inhibitor venetoclax (Venclexta). This combination is being evaluated, often alongside hypomethylating agents such as azacitidine or decitabine/cedazuridine, for the treatment of acute myeloid leukemia (AML) harboring specific genetic alterations, including KMT2A rearrangements (KMT2Ar), NPM1 mutations (NPM1m), or NUP98 rearrangements (NUP98r). By simultaneously targeting the menin-KMT2A interaction and BCL-2-mediated anti-apoptotic pathways, the combination exhibits synergistic antileukemic activity, promoting apoptosis and differentiation in leukemia cells. Clinical development is ongoing in both newly diagnosed and relapsed/refractory AML settings, including the Phase 3 EVOLVE-2 trial and the Phase 1/2 SAVE trial.
