Overview

REX-8756 is a first-in-class, oral, reversible, and highly selective small molecule inhibitor of signal transducer and activator of transcription 6 (STAT6). Developed by Recludix Pharma in partnership with Sanofi, it targets the Src Homology 2 (SH2) domain of STAT6—a critical node in the interleukin-4 (IL-4) and interleukin-13 (IL-13) signaling pathway that drives type 2 inflammatory diseases. By inhibiting STAT6 without degrading the protein, REX-8756 disrupts downstream gene transcription involved in inflammation, fibrosis, and immune dysregulation. Preclinical studies have shown complete inhibition of the STAT6 pathway both in vitro and in vivo, potent suppression of IL-4/IL-13-induced biomarkers, efficacy in models of asthma, acute lung inflammation, dermatitis (including atopic dermatitis), and favorable tolerability without evidence of hepatotoxicity or immunosuppression. IND-enabling studies are ongoing as of mid-2025[1][3][4].
