Overview

rFlu-huCTLA4 is a recombinant, chimeric oncolytic influenza A virus engineered to express a human CTLA4 antibody. It acts as an oncolytic virus—in other words, it selectively infects and destroys cancer cells—while locally delivering a checkpoint inhibitor (anti-CTLA4 antibody) to enhance anti-tumor immune responses. In preclinical models, rFlu-huCTLA4 demonstrated the ability to selectively kill hepatocellular carcinoma cells, induce apoptosis in tumor cells, and significantly activate T cells (both CD4^+^ and CD8^+^ T cells), leading to potent anti-tumor immunity. The backbone virus is derived from the A/PR/8/34 (PR8) influenza strain. rFlu-huCTLA4 is currently being studied for its efficacy in hepatocellular carcinoma and shows potential as a novel local/regional immunotherapy approach in oncology[2].
