RFT5-SMPT-dgA is an experimental antibody-drug conjugate classified as an immunotoxin. It consists of the murine monoclonal antibody RFT5, which targets the CD25 antigen (the alpha chain of the interleukin-2 receptor), chemically linked via a sterically hindered disulfide linker (SMPT) to deglycosylated ricin A-chain (dgA), a potent ribosome-inactivating toxin. The mechanism of action involves specific binding to CD25-expressing cells—such as Hodgkin and Reed-Sternberg cells in Hodgkin's lymphoma—followed by internalization and delivery of the ricin A-chain, resulting in inhibition of protein synthesis and cell death. This agent has been evaluated in phase I clinical trials for refractory Hodgkin's lymphoma, showing partial responses and manageable toxicity primarily related to vascular leak syndrome[3][6][8].
