RfuCAR is a preclinical bispecific tandem receptor CAR-T cell therapy being developed by Celluris, a Brazilian biotechnology company. The therapy is engineered with a dual-mechanism antigen-binding domain: one domain targets a tumor-associated antigen, while the second domain is designed to sequester inhibitory molecules within the tumor microenvironment (TME). This sequestration aims to modulate the TME to enhance T-cell effector function and persistence while potentially reducing toxicities such as cytokine release syndrome (CRS) and neurotoxicity. RfuCAR also incorporates a peptide-based safety switch, allowing for the therapy to be deactivated through the administration of specific receptor epitope peptides. It is currently under investigation for the treatment of both hematological and solid malignancies.
