Overview

RFUSIN2-AML1 + donor leukocyte infusion is an investigational combination immunotherapy for the treatment of relapsed or refractory acute myeloid leukemia (AML), particularly in patients who have undergone allogeneic hematopoietic stem cell transplantation. The therapy consists of two components: - **RFUSIN2-AML1** is a therapeutic cancer vaccine made from autologous or allogeneic AML blasts that are genetically modified using a lentiviral vector to express both B7.1 (CD80), a co-stimulatory molecule, and interleukin 2 (IL-2). This modification aims to enhance the immunogenicity of the leukemia cells, promoting an anti-leukemic immune response by stimulating T-cell activation[3][1][2]. - **Donor leukocyte infusion (DLI)** involves infusing lymphocytes collected from the original stem cell transplant donor into the patient. DLI is intended to induce or augment a graft-versus-leukemia effect by providing additional immune effector cells capable of targeting residual malignant cells[6][8][10]. The combination seeks to synergistically boost anti-leukemia immunity through both active vaccination and passive cellular immunotherapy.
