Overview

RG-12525 (also known as NID 525) is a small molecule leukotriene receptor antagonist initially developed by Sanofi for the treatment of asthma and other respiratory diseases. It acts as a specific, competitive, and orally available antagonist of the peptidoleukotrienes LTC4, LTD4, and LTE4, which are key mediators of bronchoconstriction and inflammation. In addition to its primary respiratory targets, RG-12525 functions as a peroxisome proliferator-activated receptor gamma (PPAR-γ) agonist and is a potent inhibitor of the CYP3A4 enzyme. Clinical studies demonstrated that oral administration of RG-12525 effectively inhibits LTD4-induced bronchoconstriction in patients with mild asthma. Although it was also investigated for the treatment of type II diabetes, its global development has been discontinued.
