RG-70099

RG-70099 is a small-molecule dual inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO2) being developed by Curadev Pharma in partnership with Roche for cancer immunotherapy. The compound targets the kynurenine pathway, which is a key mechanism used by tumors to create an immunosuppressive microenvironment. By inhibiting both IDO1 and TDO2, RG-70099 prevents the degradation of the essential amino acid tryptophan into kynurenine metabolites that inhibit T-cell and natural killer cell function. This dual inhibition strategy is intended to provide more robust immune activation compared to selective IDO1 inhibitors, which can be bypassed by the compensatory upregulation of TDO2.
