RG6468 (also known as RO7490677) is an investigational bispecific monoclonal antibody designed as a T-cell engager (TCE) for the treatment of solid tumors. Developed by Genentech and its parent company Roche, the molecule is engineered to simultaneously target Claudin 6 (CLDN6) and the CD3 receptor on T cells. Claudin 6 is a tumor-associated antigen that is highly expressed in various solid malignancies—including ovarian, lung, and testicular cancers—but has minimal expression in healthy adult tissues, making it an ideal target for selective therapy. By bridging CLDN6-expressing cancer cells with cytotoxic T cells, RG6468 facilitates the formation of an immunological synapse, leading to T-cell activation and the subsequent release of perforins and granzymes that induce tumor cell lysis. The drug is currently in Phase 1 clinical development to evaluate its safety, pharmacokinetics, and preliminary anti-tumor activity in patients with advanced solid tumors.
