RG6538

RG6538 (also known as RO7616789) is an investigational bispecific monoclonal antibody designed as a T-cell engager for the treatment of relapsed or refractory multiple myeloma. Developed by Genentech (Roche), the molecule simultaneously targets the G protein-coupled receptor class C group 5 member D (GPRC5D), which is highly and selectively expressed on malignant plasma cells, and the CD3 epsilon subunit of the T-cell receptor complex. By binding both targets, RG6538 facilitates the recruitment and activation of cytotoxic T-cells directly to the site of the myeloma cells, leading to the formation of an immunological synapse and subsequent T-cell mediated lysis of the cancer cells. It is currently being evaluated in Phase 1 clinical trials as a subcutaneous injection.
