RG7203 is a small molecule inhibitor of **phosphodiesterase 10A (PDE10A)**, an enzyme highly expressed in the striatum that modulates both **dopamine D2- and D1-dependent signaling**. By inhibiting PDE10A, RG7203 suppresses D2-mediated signaling (similar to D2 antagonists), and enhances D1-dependent signaling, which is implicated in reward-based learning. Developed primarily as an adjunctive treatment for negative symptoms in **schizophrenia**, RG7203 was intended to improve reward functioning and motivation deficits. However, clinical studies found that while RG7203 increased certain imaging markers of reward expectation, it paradoxically decreased effortful reward-based behavior and failed to demonstrate therapeutic benefit for negative symptoms in schizophrenia. The compound was discontinued for this indication[1][2][3][4][5][7].
