RG7345 (also known as RO6926496) is a humanized monoclonal antibody developed as an immunotherapy targeting the tau phosphoepitope at serine 422 (pS422), a pathological form of tau protein implicated in Alzheimer's disease. By binding to this specific epitope, which is prominent in neuronal dendrites and associated with abnormal tau relocalization and aggregation, the antibody was designed to reduce insoluble tau levels and potentially improve cognitive function. Preclinical studies demonstrated decreased brain insoluble tau and cognitive improvements in animal models. A Phase 1 clinical trial was initiated to assess safety and pharmacokinetics in healthy volunteers; however, development was discontinued during this early phase[1][2][7].
