Overview

RG7356 is a recombinant humanized monoclonal antibody of the IgG1 subclass that specifically targets the constant region of CD44, a transmembrane glycoprotein overexpressed in various malignancies, including solid tumors and hematologic cancers. By blocking CD44 and hyaluronic acid interactions, RG7356 inhibits cell adhesion and has been associated with macrophage activation in preclinical models. The drug was evaluated in phase I trials for both advanced CD44-expressing solid tumors and relapsed/refractory acute myeloid leukemia (AML). It demonstrated favorable tolerability and modest clinical activity as monotherapy, with suggestions for further research in combination regimens or settings of minimal residual disease. RG7356 was developed by Roche and has not advanced beyond early clinical development for either indication.[1][2][5][6][3]
