Overview

RG7827 is a recombinant antibody fusion protein comprising a split trimeric human 4-1BB ligand (4-1BBL), a fibroblast activation protein (FAP)-targeting Fab moiety, and an engineered Fc region to minimize interaction with FcγRs and C1q. It selectively activates the 4-1BB/CD137 immunoreceptor in FAP-positive environments, thereby aiming to generate localized T cell costimulation and antitumor immunity in malignant tissues. RG7827 was primarily developed for cancer indications such as solid tumors, metastatic colorectal cancer, urogenital cancers, and non-small cell lung cancer. Development has largely been discontinued, with remaining early-stage studies.<br>[1][2][3][4][5][9]
