Overview

RGDCRAdCOX2F is a tropism-modified conditionally replicative adenovirus vector being investigated in clinical trials for pancreatic adenocarcinoma. It's currently in Phase I clinical trials where it's administered as a direct tumor injection to evaluate safety and determine the maximum tolerated dose[1][2][5]. RGDCRAdCOX2F is a conditionally replicative adenovirus (CRAd) that has been modified with an RGD motif to enhance its binding to integrins expressed on tumor cells[3]. The virus is designed to selectively replicate in cancer cells that express cyclooxygenase-2 (COX-2), as it contains a COX-2 promoter that controls viral replication[4]. This selective targeting mechanism allows the virus to preferentially infect and destroy cancer cells while sparing normal tissues. The "RGD" in the name refers to the Arg-Gly-Asp motif that enhances viral binding to integrins on tumor cells, "CRAd" stands for conditionally replicative adenovirus, "COX2" refers to the cyclooxygenase-2 promoter that controls viral replication, and "F" likely indicates the direction of the E1 expression cassette (left to right)[4].
