RGE-80 is a small molecule drug candidate that was investigated for its potential therapeutic effects in the central nervous system, specifically targeting GABAergic neurotransmission. Developed by the Japanese pharmaceutical company Mitsubishi Tanabe Pharma, RGE-80 acts as a modulator of the GABA-A receptor complex. The primary focus of its development was for the treatment of anxiety disorders and potentially other conditions related to neuronal over-excitation. Mechanistically, it enhances the inhibitory effects of gamma-aminobutyric acid (GABA), the primary inhibitory neurotransmitter in the mammalian brain, by binding to specific sites on the GABA-A receptor. This modulation leads to an increase in chloride ion conductance, resulting in hyperpolarization of the postsynaptic neuron and a subsequent reduction in neuronal excitability. Despite its pharmacological potential, clinical development of RGE-80 appears to have been discontinued or stalled in early phases.
