RGFP-966 is a potent and highly selective small molecule inhibitor of histone deacetylase 3 (HDAC3), with an IC50 of approximately 80 nM and over 200-fold selectivity over other HDAC isoforms. Originally developed by Repligen Corporation, it serves as a critical research tool for investigating the epigenetic regulation of gene expression and protein function. In preclinical models of respiratory disease, RGFP-966 has been shown to modulate the PIM1-HDAC3-IL-33 axis; by inhibiting HDAC3, it prevents the deacetylation of IL-33, thereby reducing its secretion from lung epithelial cells and attenuating allergic airway inflammation. Beyond its role in inflammation, RGFP-966 is extensively studied for its ability to enhance synaptic plasticity and long-term memory consolidation, as well as its potential therapeutic effects in neurodegenerative conditions like Huntington's disease and various oncology settings.
