RGH-478 is an investigational, orally bioavailable small molecule developed by Gedeon Richter for the treatment of inflammatory pain conditions, such as osteoarthritis and rheumatoid arthritis. It functions as a potent and selective antagonist of the P2X3 receptor (P2X purinoceptor 3), a ligand-gated ion channel found on primary sensory neurons that mediates pain signaling in response to extracellular ATP. By inhibiting these receptors, RGH-478 is intended to reduce peripheral sensitization and chronic pain. The compound has been studied in Phase 1 trials for safety and pharmacokinetics and progressed to a Phase 2 exploratory trial (NCT04141410) in Hungary, where it was evaluated against placebo and celecoxib in patients with moderate to severe pain from osteoarthritis of the knee.
