Overview

RGT-490 is an investigational, orally bioavailable small molecule inhibitor developed by Regor Pharmaceuticals. It is designed to target the phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit alpha (PIK3CA), a key component of the PI3K/AKT/mTOR signaling pathway that is frequently mutated in various malignancies. By inhibiting mutated PIK3CA, RGT-490 aims to suppress the aberrant signaling that drives tumor cell proliferation and survival. It is currently under clinical investigation in Phase 1/1b trials for patients with locally advanced or metastatic PIK3CA-mutated solid tumors, including hormone receptor-positive, HER2-negative (HR+/HER2-) breast cancer.
