Overview

RGV005 is an engineered oncolytic vaccinia virus designed to deliver a CD19-targeted chimeric antigen receptor (CAR) gene directly to the tumor site. Developed by the Second Affiliated Hospital, School of Medicine, Zhejiang University, this therapy is intended for patients with refractory or relapsed B-cell lymphoma. The mechanism involves the *in situ* transduction of host immune cells, such as T cells and macrophages, to express CD19-CARs, effectively creating CAR-T and CAR-M cells within the tumor microenvironment. This approach combines the direct cell-lysing capabilities of an oncolytic virus with the targeted immune response of CAR-based therapy, potentially improving efficacy and safety compared to traditional systemic CAR-T cell treatments.
