Overview
**Rh-YBS** is an experimental oral colonic submucosa-targeted drug-probiotic co-delivery system for ulcerative colitis. It co-encapsulates the small molecule rhein and live *Bacillus subtilis* within yeast cell wall microparticles. The carrier is designed to target colonic microfold cells and deliver the payload to the colonic submucosa, where rhein promotes *B. subtilis* colonization. In a dextran sulfate sodium-induced mouse model of ulcerative colitis, the system increased calcitonin gene-related peptide-associated signaling, promoted goblet-cell mucus secretion, and supported mucosal repair and prevention of colitis.
