Overview

RH5.1 is a recombinant protein vaccine candidate targeting the blood-stage of *Plasmodium falciparum* malaria. It is based on the full-length ectodomain of the PfRH5 antigen, which is a key ligand required for the parasite's invasion of human red blood cells. The vaccine aims to induce potent antibodies that block this invasion step, thereby preventing blood-stage infection. RH5.1 is typically formulated with an adjuvant (Matrix-M or AS01B) to enhance immunogenicity. Clinical trials have demonstrated that RH5.1/Matrix-M is safe, highly immunogenic, and provides promising efficacy against clinical malaria, particularly when administered with a delayed third-dose regimen. It is being evaluated for its ability to complement existing malaria vaccines that target other stages of the parasite lifecycle, and is developed primarily by the University of Oxford and collaborators[1][3][4][6][7][9].
