Overview

RH5.2 virus-like particle is a second-generation blood-stage malaria vaccine candidate developed by the University of Oxford. It targets the *Plasmodium falciparum* Reticulocyte-binding protein Homologue 5 (RH5), a highly conserved protein essential for the parasite's invasion of human red blood cells. Unlike the first-generation RH5.1 protein vaccine, RH5.2 is displayed on a virus-like particle (VLP) platform, typically using the SpyTag/SpyCatcher conjugation system on AP205 VLPs. This multivalent display enhances the immunogenicity of the RH5 antigen, aiming to elicit high titers of neutralizing antibodies that block the interaction between RH5 and its erythrocyte receptor, Basigin (CD147), thereby preventing malaria infection and clinical disease.
