RH5.2-VLP is an experimental blood-stage malaria vaccine candidate developed by the University of Oxford. It utilizes a re-engineered and stabilized version of the Plasmodium falciparum reticulocyte-binding protein homolog 5 (RH5) antigen, known as RH5.2, which comprises the alpha-helical core of the protein. This immunogen is bioconjugated to hepatitis B surface antigen (HBsAg) virus-like particles (VLPs) using the 'plug-and-display' SpyTag-SpyCatcher platform technology. Formulated with Novavax's Matrix-M adjuvant, the vaccine is designed to elicit high titers of neutralizing antibodies that block the merozoite's invasion of red blood cells, a critical step in the malaria life cycle. Preclinical data suggests it offers superior immunogenicity compared to earlier soluble protein versions like RH5.1, and it is currently undergoing Phase 1/2 clinical evaluation.
