Overview

Rhenium-188-MN-14 is a radiopharmaceutical composed of the radioisotope rhenium-188 conjugated to MN-14, a monoclonal antibody (IgG) that targets carcinoembryonic antigen (CEA). The drug is designed for use in radioimmunotherapy of cancers expressing CEA, particularly advanced gastrointestinal cancers. The mechanism involves the specific binding of the MN-14 antibody to CEA-expressing tumor cells and subsequent delivery of cytotoxic beta radiation from rhenium‑188 directly to these cells. This targeted approach aims to maximize tumor cell kill while minimizing damage to surrounding healthy tissue. Clinical studies have demonstrated enhanced uptake in liver, spleen, and kidneys compared with iodine‑131–labeled analogs; red marrow suppression was identified as the dose-limiting toxicity. The agent has been investigated primarily for its potential in treating chemotherapy-resistant or advanced gastrointestinal malignancies[2][3][4].
