**Rheumavax** is an investigational, personalised autologous dendritic-cell immunotherapy for rheumatoid arthritis. Patient-derived dendritic cells are conditioned ex vivo with the irreversible nuclear factor kappa B inhibitor BAY 11-7082 to promote a tolerogenic phenotype, then loaded with liposomes containing four citrullinated autoantigen peptides derived from collagen type II, fibrinogen alpha chain, fibrinogen beta chain, and vimentin. A single intradermal administration was evaluated in a first-in-human Phase 1 study in HLA risk genotype-positive, anti-citrullinated peptide antibody-positive rheumatoid arthritis; treatment was well tolerated and showed biological evidence consistent with antigen-specific immune tolerance, including reduced effector T cells, an increased regulatory-to-effector T-cell ratio, and reduced disease activity in participants with active disease.
