Overview

rhFbgAα847 is a recombinant human fibrinogen (rhFGN) variant developed by the Leiden University Medical Center (LUMC). It is a biologic protein designed to support fibrin clot formation in the coagulation cascade. Unlike standard plasma-derived fibrinogen or wild-type recombinant fibrinogen (which typically has an Aα chain of 610 amino acids), this variant features an extended Aα chain of 847 amino acids. This specific length corresponds to the full-length cDNA sequence of the Aα chain before natural proteolytic processing or alternative splicing that results in the common plasma form. It is being investigated for the treatment of coagulopathy and perioperative bleeding, particularly in cardiac surgery and trauma-induced blood loss. In ex vivo studies, it has been compared to other fibrinogen products to evaluate its efficacy in restoring clot stability and strength.
