rhIL-15 + alemtuzumab is an investigational immunotherapy combination developed by the National Cancer Institute (NCI) for the treatment of relapsed or refractory T-cell malignancies, including adult T-cell leukemia (ATL), peripheral T-cell lymphoma (PTCL), cutaneous T-cell lymphoma (CTCL), and T-cell prolymphocytic leukemia (T-PLL). Recombinant human interleukin-15 (rhIL-15) is a cytokine that stimulates the proliferation and activation of natural killer (NK) cells and CD8+ T cells. Alemtuzumab is a humanized monoclonal antibody targeting CD52, a glycoprotein expressed on the surface of mature lymphocytes and most T-cell malignancies. The combination leverages IL-15 to expand the NK cell population, thereby enhancing alemtuzumab-mediated antibody-dependent cellular cytotoxicity (ADCC) against CD52-expressing tumor cells.
