rHMPV-Pa is an experimental live attenuated recombinant vaccine candidate for human metapneumovirus (HMPV). It is a chimeric virus in which the phosphoprotein (P) gene of HMPV has been replaced by the P gene from avian metapneumovirus (AMPV) subtype C, while all other proteins are derived from HMPV. This genetic modification confers host range attenuation, restricting replication and enhancing safety as a live intranasal vaccine. rHMPV-Pa was designed to induce both local and systemic immune responses against HMPV, aiming to protect infants and children from lower respiratory tract infections caused by this virus. In preclinical studies, it showed efficient replication in vitro and strong immunogenicity in animal models; however, phase I clinical trials revealed that it was overattenuated in seronegative children—the primary target population—resulting in insufficient immunogenicity[1][3][6]. The vaccine is not approved for use but serves as a benchmark for future live attenuated HMPV vaccine development.
