Overview

rHPIV2 is a recombinant form of the human parainfluenza virus type 2, engineered using reverse genetics to serve as a live-attenuated vaccine candidate or a viral vector platform. Developed primarily by the National Institute of Allergy and Infectious Diseases (NIAID), rHPIV2 is designed for intranasal administration to induce a localized mucosal immune response (IgA) as well as systemic immunity (IgG and T-cells) in the respiratory tract. The virus is typically attenuated through specific mutations in the P/V and L genes to ensure safety while maintaining sufficient replication in the upper respiratory tract to stimulate an immune response. As a vector, it has been modified to express heterologous antigens, such as the SARS-CoV-2 spike protein or respiratory syncytial virus (RSV) fusion protein, making it a versatile tool for preventing various respiratory infections.
