rHSV-1-APD1 is a recombinant oncolytic herpes simplex virus type 1 (HSV-1) engineered to express an anti-PD-1 antibody. This dual-function agent combines the direct tumor cell lysis of an oncolytic virus with local immune checkpoint inhibition by delivering anti-PD-1 antibody directly within the tumor microenvironment. The mechanism of action involves selective infection and destruction of cancer cells by HSV-1, while the expressed anti-PD-1 antibody blocks PD-1 signaling on T cells, thereby enhancing antitumor immune responses. This approach aims to overcome resistance to systemic checkpoint inhibitors and improve immunotherapy efficacy in solid tumors.
