Overview

RI-ACT™ (Rapidly Internalizing Receptor-Antibody Conjugate Technology) is a proprietary preclinical-stage bispecific antibody-drug conjugate (ADC) platform developed by NGM Biopharmaceuticals for the treatment of solid tumors. The architecture utilizes a bispecific antibody where one arm targets a tumor-associated antigen (TAA) to ensure specificity, while the second arm binds to a rapidly internalizing receptor (RIR). This dual-targeting approach is designed to exploit the high-speed endocytosis of the RIR to facilitate the efficient delivery and internalization of cytotoxic payloads into cancer cells, thereby enhancing tumor cell killing and potentially overcoming limitations of conventional ADCs that rely on slowly internalizing antigens.
