Overview

RI-OR2-TAT is a brain-penetrant, proteolytically stable retro-inverso peptide inhibitor designed for the treatment of Alzheimer's disease. It is a chimeric peptide consisting of RI-OR2 (a retro-inverted version of the Aβ(16–20) KLVFF sequence) and a retro-inverted HIV-1 TAT protein transduction domain. The peptide is designed to cross the blood-brain barrier and bind to Amyloid-beta (Aβ) monomers, oligomers, and fibrils, thereby inhibiting the formation of toxic Aβ species. Preclinical studies in mouse models have demonstrated that RI-OR2-TAT reduces amyloid plaque load, decreases neuroinflammation and oxidative stress, and promotes neurogenesis in the brain.
