Overview

Riamilovir, marketed under the brand name Triazavirin, is a synthetic guanine nucleotide analog and a member of the triazolotriazine class of non-nucleoside antiviral drugs. Developed through a collaboration between the Ural Federal University, the Russian Academy of Sciences, and Medsintez Pharmaceutical, it was originally designed to treat pandemic influenza strains such as H5N1 and H1N1. Its mechanism of action involves the inhibition of viral RNA-directed RNA polymerase, which prevents the synthesis of viral RNA and the replication of viral genomic fragments. In the context of the COVID-19 pandemic, riamilovir has been evaluated for its efficacy against SARS-CoV-2, including a Phase II/III trial conducted by the Wits Health Consortium in South Africa for hospitalized patients with mild-to-moderate disease.
