Ribavirin + interferon-beta-1a is a combination therapy that brings together two agents with distinct antiviral and immunomodulatory mechanisms. Ribavirin is a synthetic nucleoside analog with broad-spectrum antiviral activity against both DNA and RNA viruses. Its mechanisms include direct inhibition of viral replication by increasing mutation frequency in viral genomes (error catastrophe), competitive inhibition of inosine monophosphate dehydrogenase (IMPDH) leading to depletion of guanosine triphosphate pools necessary for viral RNA synthesis, and immunomodulation by shifting the host immune response toward a Th1 phenotype[5][6]. Interferon beta-1a is a recombinant form of human type I interferon used primarily as an immunomodulator in multiple sclerosis; it binds to specific cell surface receptors to decrease immune cell activity, reduce inflammation in the central nervous system, and suppress autoimmune responses[1][2]. In combination, these drugs have been investigated for synergistic effects against certain viral infections such as hepatitis C virus (HCV) and have also been explored as potential therapies for severe acute respiratory syndromes like COVID-19[4][8].
