Overview
RIBI Adjuvant System (RAS), also known as Ribi adjuvant or Sigma Adjuvant System, is an oil-in-water emulsion adjuvant used to enhance the immune response in vaccine formulations and antibody production. Developed by RIBI ImmunoChem Research and later acquired by Corixa Corporation (now part of GSK), it consists of squalene oil, Tween 80, and three key immunostimulatory components: monophosphoryl lipid A (MPL), trehalose dimycolate (TDM), and mycobacterial cell wall skeleton (CWS). The system primarily functions by activating Toll-like receptor 4 (TLR4), which triggers the production of cytokines and improves antigen uptake and presentation by dendritic cells. It was designed as a less toxic and more easily administered alternative to Freund's Complete Adjuvant, particularly for research and veterinary applications.
