Overview

Ribociclib is an oral, small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), developed for the treatment of hormone receptor-positive, human epidermal growth factor receptor 2 (HER2)-negative breast cancer. It works by selectively inhibiting CDK4 and CDK6, which are key regulators of cell cycle progression from the G1 to S phase through phosphorylation of the retinoblastoma protein (pRb). By blocking these kinases, ribociclib prevents cancer cell proliferation. Ribociclib is primarily used in combination with endocrine therapy such as aromatase inhibitors or fulvestrant for advanced or metastatic breast cancer in adults[1][3][4][5][6]. It is also being studied for use in early-stage breast cancer[1].
