Ribociclib is a small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), developed by Novartis, used as a targeted therapy for hormone receptor-positive, HER2-negative advanced or metastatic breast cancer. It works by blocking CDK4/6 proteins, which are involved in cell cycle progression, thereby slowing or stopping the growth of cancer cells. Ribociclib is administered in combination with either an aromatase inhibitor such as letrozole (Femara) or with fulvestrant (Faslodex), both of which are hormone therapies that further suppress estrogen-driven tumor growth. Letrozole is an aromatase inhibitor that reduces estrogen production, while fulvestrant is an estrogen receptor antagonist that blocks and degrades the estrogen receptor[1][2][3][4][5][6]. This combination approach has been shown to significantly improve progression-free and overall survival in patients with advanced breast cancer.
