Overview

Ribociclib + spartalizumab is an investigational combination therapy consisting of ribociclib, a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor, and spartalizumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). Ribociclib inhibits CDK4/6 to block cell cycle progression in cancer cells. Spartalizumab blocks PD-1 to enhance T-cell mediated immune responses against tumors. The combination has been studied primarily in advanced solid tumors such as hormone receptor-positive/HER2-negative metastatic breast cancer and recurrent/metastatic head and neck squamous cell carcinoma. Early-phase clinical trials have shown limited efficacy for the doublet with notable hepatotoxicity when combined with endocrine therapy[1][5]. Development was discontinued for some indications due to safety concerns.
